The church was originally named the Second Colored Baptist Church and was located on nearby Dexter Street. The church's name was officially changed to the Dexter Avenue Baptist Church in 1907. During his leadership, the congregation of the church voted to participate in the Montgomery Bus Boycott. The church also served as the headquarters of the Montgomery Improvement Association, a civil rights organization.
