The church was built on the site of the old church for San Lorenzo in Damaso, which had once been the home of the Benedictines and the University of Rome. In 1701, Pope Clement XI entrusted the church to the Confraternity of Saint Lawrence. The Confraternity was responsible for overseeing the daily operations of the church and served as the head of St. Lawrence's ministry. The current structure underwent major renovations in 1914 and is now considered one of the most beautiful churches in Rome.
